No. Tienen distintos significados y no son intercambiables.

Shortly es un adverbio cuyo significado más frecuente es ‘pronto’ o ‘en breve’:

The train will shortly be arriving at Victoria Station.

También puede significar ‘brevemente’:

The report shortly enumerated the key points raised at the meeting.

Cuando se usa para describir el tono o la actitud de una persona al hablar significa ‘ásperamente, secamente’ o ‘de manera cortante’:

You are not allowed in here. Please leave at once,’ he said, shortly.

In short, por otra parte, se usa para indicar que se está a punto de resumir algo en unas pocas palabras y de forma muy directa:
